The main difference, obviously, is the number of strings.  Where a 6 string guitar will have standard tuning, the 12 string acoustic guitar will have 2 low e, 1 a, 1 d, 1 g, 1 b, and 1 high e strings, in that order.  You will have the ability to tune this guitar into two different tunings.  There will also be a lot more tension on the neck, which can cause it to warp faster, after a couple of years, versus the 6 string guitar.

When you are considering how to play 12 string guitar, you will hopefully first be able to play the 6 string guitar.  Because there are fewer strings, it is much easier to play than the 12 string.  If you want to learn to how to bend the notes, you will not be able to do this on a 12 string.  You also need to realize that it will be more difficult to tune.  Now I am sure you are ready for the good reasons to learn how to learn to play 12 string guitar.

You will find that most musicians who use a 12 string, will have a 6 string as their main instrument.  The 12 string guitar will have a bright, rich sound that you just can't get with the 6 string.  The chords will sound richer and you will have the option of different tunings.  If you are a beginner, you can learn how to play 12 string guitar, it will just take a bit longer than the 6 string.  But think, if you learn on a 12 string, you will be able to play a 6 string with no problem.
